Controlled Neuron Growth By Patterned Protein Gradients
نویسنده
چکیده
page 126 Abstract: Neuron guidance is mainly controlled by the growth cone, located at the end of an axon. Extra-cellular components, including proteins, control the directional growth of these neurons; this allows for the possibility to manipulate extra-cellular factors to aide in the accurate rewiring of damaged neurons. The objective of this project is to create a standard platform on which to study neuron guidance by protein gradients using an electrospray system. The electrospray system is first studied using polystyrene latex (PSL) particles. This method deposits a uniform distribution of particles on a substrate and can be controlled to spray at constant velocities or accelerations. Neuron cells from extracted dorsal root ganglia (DRG) of fetal chickens are used to observe growth patterns under varying conditions, including various protein gradients and the presence of different proteins.

High-Content Neurite Development Study Using Optically Patterned Substrates
The study of neurite guidance in vitro relies on the ability to reproduce the distribution of attractive and repulsive guidance molecules normally expressed in vivo. The identification of subtle variations in the neurite response to changes in the spatial distribution of extracellular molecules can be achieved by monitoring the behavior of cells on protein gradients. To do this, automated high-...

Understanding the fundamentals of cell behaviour is imperative for designing and improving engineering strategies for regenerative medicine. By combining the precision of confocal microscopy with photochemistry, nerve growth factor (NGF) was chemically immobilized on chitosan films either in distinct areas or as concentration gradients. Using rhodamine as a proxy for NGF, a series of immobilize...

This paper describes a simple plasma-based dry etching method that enables patterned cell culture inside microfluidic devices by allowing patterning, fluidic bonding and sterilization steps to be carried out in a single step. This plasma-based dry etching method was used to pattern cell-adhesive and non-adhesive areas on the glass and polystyrene substrates. The patterned substrate was used for...
متن کاملAxon initiation and growth cone turning on bound protein gradients.
Extracellular gradients of secreted guidance factors are known to guide axon pathfinding and neuronal migration. These factors are likely to bind to cell surfaces or extracellular matrix, but whether and how they may act in bound gradients remains mostly unclear. In this study, we have developed a new technique for rapid production of stable microscopic gradients of substrate-bound proteins by ...
متن کاملGrowth cone guidance and neuron morphology on micropatterned laminin surfaces.
Neurite growth cones detect and respond to guidance cues in their local environment that determine stereotyped pathways during development and regeneration. Micropatterns of laminin (which was found to adsorb preferentially to photolithographically defined hydrophobic areas of micropatterns) were here used to model adhesive pathways that might influence neurite extension. The responses of growt...
